            <title><![CDATA[Mechanics of two filaments in tight orthogonal contact]]></title>

            <description><![CDATA[<p class="para" id="N65542">Knots, knits, and weaves have been technologically essential across civilizations, and their significance remains undiminished today. In these systems, it is challenging to understand the equilibria of the deformable filaments with their tight contacts due to the intricate geometry of touching tubular volumes of small, but nonvanishing, radius. This article considers a specific canonical context for filaments in contact: the orthogonal clasp. We quantify the significant mismatches between the physical reality of orthogonal clasps and the simplifying assumptions underpinning conventional descriptive models, such as the classic capstan equation. Nevertheless, we show that a simple, geometric model qualitatively captures the striking localization patterns in the observed contact-pressure fields.</p><p class="para" id="N65539">Networks of flexible filaments often involve regions of tight contact. Predictively understanding the equilibrium configurations of these systems is challenging due to intricate couplings between topology, geometry, large nonlinear deformations, and friction. Here, we perform an in-depth study of a simple, yet canonical, problem that captures the essence of contact between filaments. In the orthogonal clasp, two filaments are brought into contact, with each centerline lying in one of a pair of orthogonal planes. Our data from X-ray tomography (μ<div class="imageVideo"><img src="" alt=""/></div>CT) and mechanical testing experiments are in excellent agreement with finite element method (FEM) simulations. Despite the apparent simplicity of the physical system, the data exhibit strikingly unintuitive behavior, even when the contact is frictionless. Specifically, we observe a curvilinear diamond-shaped ridge in the contact-pressure field between the two filaments, sometimes with an inner gap. When a relative displacement is imposed between the filaments, friction is activated, and a highly asymmetric pressure field develops. These findings contrast to the classic capstan analysis of a single filament wrapped around a rigid body. Both the μ<div class="imageVideo"><img src="" alt=""/></div>CT and FEM data indicate that the cross-sections of the filaments can deform significantly. Nonetheless, an idealized geometrical theory assuming undeformable tube cross-sections and neglecting elasticity rationalizes our observations qualitatively and highlights the central role of the small, but nonzero, tube radius of the filaments. We believe that our orthogonal clasp analysis provides a building block for future modeling efforts in frictional contact mechanics of more complex filamentary structures.</p>]]></description>

            <title><![CDATA[Human hand as a powerless and multiplexed infrared light source for information decryption and complex signal generation]]></title>

            <description><![CDATA[<p class="para" id="N65542">The utilization of electricity-powered light sources as external stimuli is limited by the cost and energy loss. Here we demonstrate that the human hand can be used as a natural IR light source without the need of external power. As each finger can serve as an independent light source, the hand can also be utilized as a multiplexed IR light source. This work provides a different insight on using the hand in a functional system, such as the information encryption and complex signal generation systems demonstrated here, which will help move forward the effort in the integration of human components into various systems to increase the level of intelligence and achieve ultimate control of these systems.</p><p class="para" id="N65539">With the increasing pursuit of intelligent systems, the integration of human components into functional systems provides a promising route to the ultimate human-compatible intelligent systems. In this work, we explored the integration of the human hand as the powerless and multiplexed infrared (IR) light source in different functional systems. With the spontaneous IR radiation, the human hand provides a different option as an IR light source. Compared to engineered IR light sources, the human hand brings sustainability with no need of external power and also additional level of controllability to the functional systems. Besides the whole hand, each finger of the hand can also independently provide IR radiation, and the IR radiation from each finger can be selectively diffracted by specific gratings, which helps the hand serve as a multiplexed IR light source. Considering these advantages, we show that the human hand can be integrated into various engineered functional systems. The integration of hand in an encryption/decryption system enables both unclonable and multilevel information encryption/decryption. We also demonstrate the use of the hand in complex signal generation systems and its potential application in sign language recognition, which shows a simplified recognition process with a high level of accuracy and robustness. The use of the human hand as the IR light source provides an alternative sustainable solution that will not only reduce the power used but also help move forward the effort in the integration of human components into functional systems to increase the level of intelligence and achieve ultimate control of these systems.</p>]]></description>

            <title><![CDATA[Toward net-zero sustainable aviation fuel with wet waste–derived volatile fatty acids]]></title>

            <description><![CDATA[<p class="para" id="N65542">To meet the growing demand for sustainable aviation fuels (SAF), conversion pathways are needed that leverage wet waste carbon and meet jet fuel property specifications. Here, we demonstrate SAF production from food waste–derived volatile fatty acids (VFA) by targeting normal paraffins for a near-term path to market and branched isoparaffins to increase the renewable content long term. Combining these distinct paraffin structures was shown to synergistically improve VFA-SAF flash point and viscosity to increase the renewable blend limit to 70%. Life cycle analysis shows the dramatic impact on the carbon footprint if food waste is diverted from landfills to produce VFA-SAF, highlighting the potential to meet jet fuel safety, operability, and environmental goals.</p><p class="para" id="N65539">With the increasing demand for net-zero sustainable aviation fuels (SAF), new conversion technologies are needed to process waste feedstocks and meet carbon reduction and cost targets. Wet waste is a low-cost, prevalent feedstock with the energy potential to displace over 20% of US jet fuel consumption; however, its complexity and high moisture typically relegates its use to methane production from anaerobic digestion. To overcome this, methanogenesis can be arrested during fermentation to instead produce C<sub>2</sub> to C<sub>8</sub> volatile fatty acids (VFA) for catalytic upgrading to SAF. Here, we evaluate the catalytic conversion of food waste–derived VFAs to produce n-paraffin SAF for near-term use as a 10 vol% blend for ASTM “Fast Track” qualification and produce a highly branched, isoparaffin VFA-SAF to increase the renewable blend limit. VFA ketonization models assessed the carbon chain length distributions suitable for each VFA-SAF conversion pathway, and food waste–derived VFA ketonization was demonstrated for &gt;100 h of time on stream at approximately theoretical yield. Fuel property blending models and experimental testing determined normal paraffin VFA-SAF meets 10 vol% fuel specifications for “Fast Track.” Synergistic blending with isoparaffin VFA-SAF increased the blend limit to 70 vol% by addressing flashpoint and viscosity constraints, with sooting 34% lower than fossil jet. Techno-economic analysis evaluated the major catalytic process cost-drivers, determining the minimum fuel selling price as a function of VFA production costs. Life cycle analysis determined that if food waste is diverted from landfills to avoid methane emissions, VFA-SAF could enable up to 165% reduction in greenhouse gas emissions relative to fossil jet.</p>]]></description>

            <title><![CDATA[Artificial intelligence velocimetry and microaneurysm-on-a-chip for three-dimensional analysis of blood flow in physiology and disease]]></title>

            <description><![CDATA[<p class="para" id="N65542">Microfluidics is an important in vitro platform to gain insights into mechanics of blood flow and mechanisms of pathophysiology of human diseases. Extraction of 3D fields in microfluidics with dense cell suspensions remains a formidable challenge. We present artificial-intelligence velocimetry (AIV) as a general platform to determine 3D flow fields and a microaneurysm-on-a-chip to simulate blood flow in microaneurysms in patients with diabetic retinopathy. AIV is built on physics-informed neural networks that integrate seamlessly 2D images from microfluidic experiments or in vivo observations with physical laws to estimate full 3D velocity and stress fields. AIV could be integrated into imaging technologies to automatically infer key hemodynamic metrics from in vivo and in vitro biomedical images.</p><p class="para" id="N65539">Understanding the mechanics of blood flow is necessary for developing insights into mechanisms of physiology and vascular diseases in microcirculation. Given the limitations of technologies available for assessing in vivo flow fields, in vitro methods based on traditional microfluidic platforms have been developed to mimic physiological conditions. However, existing methods lack the capability to provide accurate assessment of these flow fields, particularly in vessels with complex geometries. Conventional approaches to quantify flow fields rely either on analyzing only visual images or on enforcing underlying physics without considering visualization data, which could compromise accuracy of predictions. Here, we present artificial-intelligence velocimetry (AIV) to quantify velocity and stress fields of blood flow by integrating the imaging data with underlying physics using physics-informed neural networks. We demonstrate the capability of AIV by quantifying hemodynamics in microchannels designed to mimic saccular-shaped microaneurysms (microaneurysm-on-a-chip, or MAOAC), which signify common manifestations of diabetic retinopathy, a leading cause of vision loss from blood-vessel damage in the retina in diabetic patients. We show that AIV can, without any a priori knowledge of the inlet and outlet boundary conditions, infer the two-dimensional (2D) flow fields from a sequence of 2D images of blood flow in MAOAC, but also can infer three-dimensional (3D) flow fields using only 2D images, thanks to the encoded physics laws. AIV provides a unique paradigm that seamlessly integrates images, experimental data, and underlying physics using neural networks to automatically analyze experimental data and infer key hemodynamic indicators that assess vascular injury.</p>]]></description>

            <title><![CDATA[Shape anisotropy-governed locomotion of surface microrollers on vessel-like microtopographies against physiological flows]]></title>

            <description><![CDATA[<p class="para" id="N65542">Controlled microrobotic navigation in blood vessels holds significant potential to revolutionize targeted drug delivery. Navigation on the surface of the blood vessels is advantageous because of decreased flow velocities; however, surface microtopography of blood vessels, in the size scale of the microrobots, is a major hurdle for robust locomotion of surface-rolling microrobots against the blood flow. Here, we show the effect of the body-shape anisotropy of the microrollers on their locomotion capability over vessel-like microtopographies. We demonstrate by experiments and simulations that the microrollers with slender bodies are more robust to locomote on biological microtopographies due to their favorable hydrodynamic interactions. Thus, such anisotropically shaped microrollers would be more viable and robust in future in vivo medical applications.</p><p class="para" id="N65539">Surface microrollers are promising microrobotic systems for controlled navigation in the circulatory system thanks to their fast speeds and decreased flow velocities at the vessel walls. While surface propulsion on the vessel walls helps minimize the effect of strong fluidic forces, three-dimensional (3D) surface microtopography, comparable to the size scale of a microrobot, due to cellular morphology and organization emerges as a major challenge. Here, we show that microroller shape anisotropy determines the surface locomotion capability of microrollers on vessel-like 3D surface microtopographies against physiological flow conditions. The isotropic (single, 8.5 µm diameter spherical particle) and anisotropic (doublet, two 4 µm diameter spherical particle chain) magnetic microrollers generated similar translational velocities on flat surfaces, whereas the isotropic microrollers failed to translate on most of the 3D-printed vessel-like microtopographies. The computational fluid dynamics analyses revealed larger flow fields generated around isotropic microrollers causing larger resistive forces near the microtopographies, in comparison to anisotropic microrollers, and impairing their translation. The superior surface-rolling capability of the anisotropic doublet microrollers on microtopographical surfaces against the fluid flow was further validated in a vessel-on-a-chip system mimicking microvasculature. The findings reported here establish the design principles of surface microrollers for robust locomotion on vessel walls against physiological flows.</p>]]></description>

            <title><![CDATA[T cells selectively filter oscillatory signals on the minutes timescale]]></title>

            <description><![CDATA[<p class="para" id="N65542">Immune cells in the body encounter stimuli in complex temporal patterns, but our understanding of how the timing of stimulation determines immune responses is incomplete. We engineered T cells obtained from healthy human donors to respond to light as a stimulus in a laboratory setting. We discovered that T cells can filter out specific signals based only on signal frequency by stimulating engineered T cells with light-based temporal stimulation patterns designed to mimic the dynamic patterns a natural T cell might experience in the human body. This filtering mechanism could contribute to how immune cells distinguish between foreign/altered cells versus self-cells in vivo<i>,</i> since foreign/altered and self-cells exhibit highly distinct interaction dynamics with T cells within the body.</p><p class="para" id="N65539">T cells experience complex temporal patterns of stimulus via receptor–ligand-binding interactions with surrounding cells. From these temporal patterns, T cells are able to pick out antigenic signals while establishing self-tolerance. Although features such as duration of antigen binding have been examined, our understanding of how T cells interpret signals with different frequencies or temporal stimulation patterns is relatively unexplored. We engineered T cells to respond to light as a stimulus by building an optogenetically controlled chimeric antigen receptor (optoCAR). We discovered that T cells respond to minute-scale oscillations of activation signal by stimulating optoCAR T cells with tunable pulse trains of light. Systematically scanning signal oscillation period from 1 to 150 min revealed that expression of CD69, a T cell activation marker, reached a local minimum at a period of ∼25 min (corresponding to 5 to 15 min pulse widths). A combination of inhibitors and genetic knockouts suggest that this frequency filtering mechanism lies downstream of the Erk signaling branch of the T cell response network and may involve a negative feedback loop that diminishes Erk activity. The timescale of CD69 filtering corresponds with the duration of T cell encounters with self-peptide–presenting APCs observed via intravital imaging in mice, indicating a potential functional role for temporal filtering in vivo. This study illustrates that the T cell signaling machinery is tuned to temporally filter and interpret time-variant input signals in discriminatory ways.</p>]]></description>

            <title><![CDATA[Neural interfacing architecture enables enhanced motor control and residual limb functionality postamputation]]></title>

            <description><![CDATA[<p class="para" id="N65542">Despite advancements in prosthetic technologies, persons with amputation today suffer great diminution in mobility and quality of life. This is largely due to an outdated amputation paradigm that precludes efficacious communication between the residual limb and prosthesis. An amputation method utilizing agonist–antagonist myoneural interfaces (AMIs) constructs neuromuscular substrates in the residual limb to avail enhanced sensorimotor signaling. In our study, subjects with AMI amputation demonstrate improved motor control, phantom sensations, range of motion, and decreased pain when compared to patients with traditional amputation. With the demonstrated increases in motor coordination and position differentiation, our results suggest that patients with AMI amputation will be able to more efficaciously control bionic prostheses.</p><p class="para" id="N65539">Despite advancements in prosthetic technologies, patients with amputation today suffer great diminution in mobility and quality of life. We have developed a modified below-knee amputation (BKA) procedure that incorporates agonist–antagonist myoneural interfaces (AMIs), which surgically preserve and couple agonist–antagonist muscle pairs for the subtalar and ankle joints. AMIs are designed to restore physiological neuromuscular dynamics, enable bidirectional neural signaling, and offer greater neuroprosthetic controllability compared to traditional amputation techniques. In this prospective, nonrandomized, unmasked study design, 15 subjects with AMI below-knee amputation (AB) were matched with 7 subjects who underwent a traditional below-knee amputation (TB). AB subjects demonstrated significantly greater control of their residual limb musculature, production of more differentiable efferent control signals, and greater precision of movement compared to TB subjects (<i>P</i> &lt; 0.008). This may be due to the presence of greater proprioceptive inputs facilitated by the significantly higher fascicle strains resulting from coordinated muscle excursion in AB subjects (<i>P</i> &lt; 0.05). AB subjects reported significantly greater phantom range of motion postamputation (AB: 12.47 ± 2.41, TB: 10.14 ± 1.45 degrees) when compared to TB subjects (<i>P</i> &lt; 0.05). Furthermore, AB subjects also reported less pain (12.25 ± 5.37) than TB subjects (17.29 ± 10.22) and a significant reduction when compared to their preoperative baseline (<i>P</i> &lt; 0.05). Compared with traditional amputation, the construction of AMIs during amputation confers the benefits of enhanced physiological neuromuscular dynamics, proprioception, and phantom limb perception. Subjects’ activation of the AMIs produces more differentiable electromyography (EMG) for myoelectric prosthesis control and demonstrates more positive clinical outcomes.</p>]]></description>

            <description><![CDATA[<p class="para" id="N65542">We demonstrate an approach to generate a new type of X-ray imaging mode, which is called omnidirectional X-ray differential phase imaging. The proposed method enables us to detect the subtle phase changes in all directions of the imaging plane, which complements conventional X-ray imaging methods with information that they cannot provide. Importantly, the omnidirectional dark-field images can also be simultaneously retrieved for studying a wide range of complicated samples, particularly strongly ordered systems. The extracted information will not only provide insights into the microarchitecture of materials, but also enrich our understanding the macroscopic behavior. The presented technique could potentially open up numerous practical imaging applications in both biomedical research and materials science.</p><p class="para" id="N65539">Ever since the discovery of X-rays, tremendous efforts have been made to develop new imaging techniques for unlocking the hidden secrets of our world and enriching our understanding of it. X-ray differential phase contrast imaging, which measures the gradient of a sample’s phase shift, can reveal more detail in a weakly absorbing sample than conventional absorption contrast. However, normally only the gradient’s component in two mutually orthogonal directions is measurable. In this article, omnidirectional differential phase images, which record the gradient of phase shifts in all directions of the imaging plane, are efficiently generated by scanning an easily obtainable, randomly structured modulator along a spiral path. The retrieved amplitude and main orientation images for differential phase yield more information than the existing imaging methods. Importantly, the omnidirectional dark-field images can be simultaneously extracted to study strongly ordered scattering structures. The proposed method can open up new possibilities for studying a wide range of complicated samples composed of both heavy, strongly scattering atoms and light, weakly scattering atoms.</p>]]></description>

            <title><![CDATA[Exhaled aerosol increases with COVID-19 infection, age, and obesity]]></title>

            <description><![CDATA[<p class="para" id="N65542">Superspreading events have distinguished the COVID-19 pandemic from the early outbreak of the disease. Our studies of exhaled aerosol suggest that a critical factor in these and other transmission events is the propensity of certain individuals to exhale large numbers of small respiratory droplets. Our findings indicate that the capacity of airway lining mucus to resist breakup on breathing varies significantly between individuals, with a trend to increasing with the advance of COVID-19 infection and body mass index multiplied by age (i.e., BMI-years). Understanding the source and variance of respiratory droplet generation, and controlling it via the stabilization of airway lining mucus surfaces, may lead to effective approaches to reducing COVID-19 infection and transmission.</p><p class="para" id="N65539">COVID-19 transmits by droplets generated from surfaces of airway mucus during processes of respiration within hosts infected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus. We studied respiratory droplet generation and exhalation in human and nonhuman primate subjects with and without COVID-19 infection to explore whether SARS-CoV-2 infection, and other changes in physiological state, translate into observable evolution of numbers and sizes of exhaled respiratory droplets in healthy and diseased subjects. In our observational cohort study of the exhaled breath particles of 194 healthy human subjects, and in our experimental infection study of eight nonhuman primates infected, by aerosol, with SARS-CoV-2, we found that exhaled aerosol particles vary between subjects by three orders of magnitude, with exhaled respiratory droplet number increasing with degree of COVID-19 infection and elevated BMI-years. We observed that 18% of human subjects (35) accounted for 80% of the exhaled bioaerosol of the group (194), reflecting a superspreader distribution of bioaerosol analogous to a classical 20:80 superspreader of infection distribution. These findings suggest that quantitative assessment and control of exhaled aerosol may be critical to slowing the airborne spread of COVID-19 in the absence of an effective and widely disseminated vaccine.</p>]]></description>

            <title><![CDATA[Exploring the property space of periodic cellular structures based on crystal networks]]></title>

            <description><![CDATA[<p class="para" id="N65542">Finding genuine novelty in cellular structures is inherently difficult due to the numerous possible topological and geometrical configurations and their complex mechanical and physical interrelations. Here, we draw inspiration from the incredibly rich collection of crystallographic periodic networks that we interpret from a structural point of view to identify and design novel cellular structures with unique properties. We provide a ready-to-use catalog with more than 17,000 unique entries and show how crystallographic symmetries relate to their mechanical properties. Our work provides a foundation to support future applications in science and engineering, ranging from mechanical and optical metamaterials, over bone tissue engineering, to the design of electrochemical devices.</p><p class="para" id="N65539">The properties of periodic cellular structures strongly depend on the regular spatial arrangement of their constituent base materials and can be controlled by changing the topology and geometry of the repeating unit cell. Recent advances in three-dimensional (3D) fabrication technologies more and more expand the limits of fabricable real-world architected materials and strengthen the need of novel microstructural topologies for applications across all length scales and fields in both fundamental science and engineering practice. Here, we systematically explore, interpret, and analyze publicly available crystallographic network topologies from a structural point of view and provide a ready-to-use unit cell catalog with more than 17,000 unique entries in total. We show that molecular crystal networks with atoms connected by chemical bonds can be interpreted as cellular structures with nodes connected by mechanical bars. By this, we identify new structures with extremal properties as well as known structures such as the octet-truss or the Kelvin cell and show how crystallographic symmetries are related to the mechanical properties of the structures. Our work provides inspiration for the discovery of novel cellular structures and paves the way for computational methods to explore and design microstructures with unprecedented properties, bridging the gap between microscopic crystal chemistry and macroscopic structural engineering.</p>]]></description>

            <title><![CDATA[Wireless, soft electronics for rapid, multisensor measurements of hydration levels in healthy and diseased skin]]></title>

            <description><![CDATA[<p class="para" id="N65542">Wireless electronics for monitoring of skin hydration in a quantitative fashion have broad relevance to our understanding of dermatological health and skin structure in both clinical and home settings. Here, we present a miniaturized, long-range automated system that adheres gently to the skin to yield quantitative recordings of skin water content for both epidermis and dermis. This system supports capabilities in characterizing skin barrier, assessing severity of skin diseases, and evaluating cosmetic and medication efficacy, with high levels of repeatability and insensitivity to ambient. Benchtop and pilot studies on patients with skin diseases highlight key features of these devices and their potential for broad utility in clinical research and in home settings to guide the management of disorders of the skin.</p><p class="para" id="N65539">Precise, quantitative measurements of the hydration status of skin can yield important insights into dermatological health and skin structure and function, with additional relevance to essential processes of thermoregulation and other features of basic physiology. Existing tools for determining skin water content exploit surrogate electrical assessments performed with bulky, rigid, and expensive instruments that are difficult to use in a repeatable manner. Recent alternatives exploit thermal measurements using soft wireless devices that adhere gently and noninvasively to the surface of the skin, but with limited operating range (∼1 cm) and high sensitivity to subtle environmental fluctuations. This paper introduces a set of ideas and technologies that overcome these drawbacks to enable high-speed, robust, long-range automated measurements of thermal transport properties via a miniaturized, multisensor module controlled by a long-range (∼10 m) Bluetooth Low Energy system on a chip, with a graphical user interface to standard smartphones. Soft contact to the surface of the skin, with almost zero user burden, yields recordings that can be quantitatively connected to hydration levels of both the epidermis and dermis, using computational modeling techniques, with high levels of repeatability and insensitivity to ambient fluctuations in temperature. Systematic studies of polymers in layered configurations similar to those of human skin, of porcine skin with known levels of hydration, and of human subjects with benchmarks against clinical devices validate the measurement approach and associated sensor hardware. The results support capabilities in characterizing skin barrier function, assessing severity of skin diseases, and evaluating cosmetic and medication efficacy, for use in the clinic or in the home.</p>]]></description>
